Absolutely include a cover letter. Without a cover letter, people will assume you just don’t care about the job, and you’re just firing you resume off to everyone in town. (Which you are, but there’s no need to remind them of that.)
I write the cover letter in the actual email, unless otherwise specified. But because things can get separated when printed out, don’t forget to include your phone number in your email. You don’t want them putting your email in the “call back” file, and then not have a number to reach you.
